Life's Engines: How Microbes Made Earth Habitable
For almost four billion years, microbes had the primordial oceans all to themselves. The stewards of Earth, these organisms transformed the chemistry of our planet to make it habitable for plants, animals, and us. Life's Engines takes readers deep into the microscopic world to explore how these marvelous creatures made life on Earth possible--and how human life today would cease to exist without them. Paul Falkowski looks under the hood of microbes to find the engines of life, the actual working parts that do the biochemical heavy lifting for every living organism on Earth. With insight and humor, he explains how these miniature engines are built--and how they have been appropriated by and assembled like Lego sets within every creature that walks, swims, or flies. Falkowski shows how evolution works to maintain this core machinery of life, and how we and other animals are veritable conglomerations of microbes. A vibrantly entertaining book about the microbes that support our very existence, Life's Engines will inspire wonder about these elegantly complex nanomachines that have driven life since its origin. It also issues a timely warning about the dangers of tinkering with that machinery to make it more efficient at meeting the ever-growing demands of humans in the coming century.

Oxygen: The Molecule That Made the World
Oxygen has had extraordinary effects on life. Three hundred million years ago, in Carboniferous times, dragonflies grew as big as seagulls, with wingspans of nearly a metre. Researchers claim they could have flown only if the air had contained more oxygen than today - probably as much as 35 per cent. Giant spiders, tree-ferns, marine rock formations and fossil charcoals all tell the same story. High oxygen levels may also explain the global firestorm that contributed to the demise of the dinosaurs after the asteroid impact. The strange and profound effects that oxygen has had on the evolution of life pose a riddle, which this book sets out to answer. Oxygen is a toxic gas. Divers breathing pure oxygen at depth suffer from convulsions and lung injury. Fruit flies raised at twice normal atmospheric levels of oxygen live half as long as their siblings. Reactive forms of oxygen, known as free radicals, are thought to cause ageing in people. Yet if atmospheric oxygen reached 35 per cent in the Carboniferous, why did it promote exuberant growth, instead of rapid ageing and death? Oxygen takes the reader on an enthralling journey, as gripping as a thriller, as it unravels the unexpected ways in which oxygen spurred the evolution of life and death. The book explains far more than the size of ancient insects: it shows how oxygen underpins the origin of biological complexity, the birth of photosynthesis, the sudden evolution of animals, the need for two sexes, the accelerated ageing of cloned animals like Dolly the sheep, and the surprisingly long lives of bats and birds. Drawing on this grand evolutionary canvas, Oxygen offers fresh perspectives on our own lives and deaths, explaining modern killer diseases, why we age, and what we can do about it. Advancing revelatory new ideas, following chains of evidence, the book ranges through many disciplines, from environmental sciences to molecular medicine. The result is a captivating vision of contemporary science and a humane synthesis of our place in nature. This remarkable book will redefine the way we think about the world.

Exploring Personal Genomics
Rapid advances in high-throughput genome sequencing technologies foreshadow a near-future in which millions of individuals will gain affordable access to their complete genome sequence. This promises to offer unprecedented insights into the fundamental biological nature of ourselves and our species: where we came from, how we begin our lives, how we develop and grow, how we interact with our environment, how we get sick, how we get well, and how we age. Personal genomics is an essential component of the inevitable transition towards personalized health and medicine. As the medical establishment begins to explore and evaluate the role of personal genomics in health and medicine, both clinicians and patients alike will gain from becoming well versed in both the power and the pitfalls of personal genomic information. Furthermore, it is likely that all students of the biomedical sciences will soon be required to gain crucial understanding in the emerging field of personal genomics. Exploring Personal Genomics provides a novel, inquiry-based approach to the understanding and interpretation of the practical, medical, physiological, and societal aspects of personal genomic information. The material is presented in two parts: the first provides readers of all backgrounds with a fundamental understanding of the biology of human genomes, information on how to obtain and understand digital representations of personal genomic data, tools and techniques for exploring the personal genomics of ancestry and genealogy, discovery and interpretation of genetic trait associations, and the role of personal genomics in drug response. The second part offers more advanced readers an understanding of the science, tools, and techniques for investigating interactions between a personal genome and the environment, connecting DNA to physiology, and assessing rare variants and structural variation. This book aims to support undergraduate and graduate studies in medicine, genetics, molecular biology, and bioinformatics. Additionally, the design of the content is such that medical practitioners, professionals working in the biomedical sciences or related fields, and motivated lay individuals interested in exploring their personal genetic data should find it relevant and approachable.

Molecular Cloning: A Laboratory Manual
Molecular Cloning has served as the foundation of technical expertise in labs worldwide for 30 years. No other manual has been so popular, or so influential. Molecular Cloning, Fourth Edition, by the celebrated founding author Joe Sambrook and new co-author, the distinguished HHMI investigator Michael Green, preserves the highly praised detail and clarity of previous editions and includes specific chapters and protocols commissioned for the book from expert practitioners at Yale, U Mass, Rockefeller University, Texas Tech, Cold Spring Harbor Laboratory, Washington University, and other leading institutions. The theoretical and historical underpinnings of techniques are prominent features of the presentation throughout, information that does much to help trouble-shoot experimental problems. For the fourth edition of this classic work, the content has been entirely recast to include nucleic-acid based methods selected as the most widely used and valuable in molecular and cellular biology laboratories. Core chapters from the third edition have been revised to feature current strategies and approaches to the preparation and cloning of nucleic acids, gene transfer, and expression analysis. They are augmented by 12 new chapters which show how DNA, RNA, and proteins should be prepared, evaluated, and manipulated, and how data generation and analysis can be handled. The new content includes methods for studying interactions between cellular components, such as microarrays, next-generation sequencing technologies, RNA interference, and epigenetic analysis using DNA methylation techniques and chromatin immunoprecipitation. To make sense of the wealth of data produced by these techniques, a bioinformatics chapter describes the use of analytical tools for comparing sequences of genes and proteins and identifying common expression patterns among sets of genes. Please take a look at our book preview site at molecularcloning.com. Building on thirty years of trust, reliability, and authority, the fourth edition of Molecular Cloning is the new gold standard--the one indispensable molecular biology laboratory manual and reference source.

Introduction to Protein Science: Architecture, Function, and Genomics
Proteins are essential to life, having a great variety of roles in all organisms. They are the ultimate micro-machines: some are building blocks, joining with other substances to make the cells from which we are formed; some are catalysts, speeding up biochemical reactions to keep our cells active and alive; yet others help cells to communicate, to move, to build up the complex mix of tissues that make up our bodies, and to regulate unfolding programmes of development. Introduction to Protein Science provides a broad ranging introduction to the contemporary study of proteins in health and disease, suitable for students on biological, biochemical, and biomedical degrees internationally. It relates the study of proteins to the context of modern high-throughput data streams of genomics and proteomics. Describing basic principles of protein structure and methods for studying them, the book illustrates the wide variety of functions that proteins have, showing how the structures of proteins are intimately linked to their functions. Building on the success of the first edition, the new edition integrates treatment of bioinformatics, databases and computational methods of determining and predicting protein structure and function, and demonstrates how these methods are paving the way for the ability to design novel proteins with specific desired characteristics. With added coverage of recent developments in the field, particularly in high-throughput methods; a new chapter on enzyme kinetics; extended coverage of topics such as protein evolution, and the immune system; as well as the inclusion of new end-of-chapter exercises, problems and weblems, the book captures the current state of excitement in a way that all biosciences students and instructors will find appealing. Biophysics of DNA-Protein Interactions: From Single Molecules to Biological Systems
Depite the rapid expansion of the field of biophysics, there are very few books that comprehensively treat specific topics in this area. Recently, the field of single molecule biophysics has developed very quickly, and a few books specifically treating single molecule methods are beginning to appear. However, the promise of single molecule biophysics is to contribute to the understanding of specific fields of biology using new methods. This book would focus on the specific topic of the biophysics of DNA-protein interactions, and would include the use of new approaches, including both bulk methods as well as single molecule methods. This would make the book attractive to anyone working in the general area of DNA-protein interactions, which is of course a much wider market than just single molecule biophysicists or even biophysicists. The subject of the book will be the biophysics of DNA-protein interactions, and will include new methods and results that describe the physical mechanism by which proteins interact with DNA. For example, there has been much recent work on the mechanism by which proteins search for specific binding sites on DNA. A few chapters will be devoted to experiments and theory that shed light on this important problem. We will also cover proteins that alter DNA properties to facilitate interactions important for transcription or replication. Another section of the book will cover the biophysical mechanism by which motor proteins interact with DNA. Finally, we will cover larger protein-DNA complexes, such as replication forks, recombination complexes, DNA repair interactions, and their chromatin context.

Behavioral Pediatrics: Research and Practice
As in any field of clinical or scientific endeavor, a cataloguing of the techniques and findings of behavioral pediatrics must follow shortly after the first few years of systematic work. This book has been designed to serve this initial summary function for the field. It represents a first attempt to bring together in one place the definition and scope of be havioral pediatrics and to outline current research and treatment ap proaches to various organic disorders, clinical settings, and problem areas in which a sufficient body of knowledge has accrued for author itative statement. As the first text in a rapidly expanding area, our decisions regarding the topics to be covered and the contributors to this volume were guided by our desire to represent what we would consider to exemplify the field in its early development: pragmatic and thorough study of signif icant problems from a base of sound scientific inquiry. Each of the topics addressed in the present volume develops, in a preliminary fashion, an epistemology for current practice and future study. All of the contrib uting authors have been involved in the development of their specialty areas through their research and, importantly, through their clinical work in the hospitals affiliated with the medical schools in which they hold appointments.

Microorganisms to Combat Pollution
This volume contains material first presented at an international workshop on the 'Use of Microorganisms to Combat Pollution', held in Israel, May 10--18, 1992. The workshop was sponsored by the Bat-Sheva de Rothschild Foundation for the Advancement of Science and included microbiologists, biochemists and geneticists from universities, environmental agencies and the military. Each of the contributors to this volume is an ack nowledged expert on the treatment of one or more types of pollution using microorganisms or their enzymes. This book differs from most published symposia proceedings in the breadth of coverage of each subject. Most of the chapters are divided into three parts: (a) A general presentation of the source and toxicity of the pollutant, (b) a review of the current state-of-the-science on the biodegradation of that pollutant and (c) the authors' unique research experiences on the problem. In several examples, the authors have presented data from both laboratory studies and field trials. Thus, the book contains not only the theoretical background on the biodegradation of pollutants, but also practical experiences in applying this knowledge to solving significant pollution problems.

Aphasia and Brain Organization
This book presents the work on aphasia coming out of the Institute for Aphasia and Stroke in Norway during its 10 years of existence. Rather than reviewing previously presented work, it was my desire to give a unified analysis and discussion of our accumulated data. The empirical basis for the analysis is a fairly large group (249 patients) investigated with a standard, comprehensive set of procedures. Tests of language functions must be developed anew for each language, but comparison of my findings with other recent compre hensive studies of aphasia is faciliated by close parallels in test meth ods (Chapter 2). The classification system used is currently the most accepted neurological system, but I have operationalized it for research purposes (Chapter 3). The analyses presented are based on the view that aphasia is an aspect of a multidimensional disturbance of brain function. Find ings of associated disturbances and variations in the aphasic condition over time have been dismissed by some as irrelevant to the study of aphasia as a language deficit. My view is that this rich and complex set of findings gives important clues to the organization of brain functions in humans. I present analyses of the relationship of aphasia to neuropsychological disorders in conceptual organization, memory, visuospatial abilities and apraxia (Chapters 4, 5, and 6), and I study the variations with time of the aphasic condition (Chapter 8).

The Violinists Thumb: And Other Extraordinary True Stories as Written by Our DNA
Did the human race almost go extinct? Can genetics explain a cat lady's love for felines? How does DNA lead to people with no fingerprints or humans born with tails? And how did the right combination of genes create the exceptionally flexible thumbs and fingers of a truly singular violinist? Unravelling the genetic code hasn't always been easy - from its earliest days, genetics has been rife with infighting, backstabbing and controversial theories - but scientists can now finally read the astounding stories inscribed in our DNA. As we make advances into DNA mapping and modification, genetics will continue to be the hottest topic in science, shaping the very make-up of our bodies and the world around us. With the same masterful combination of science, history and culture he brought to The Disappearing Spoon , Sam Kean untangles the secrets of our genetic code, explaining how genetics has shaped our past and how DNA will determine humankind's future.

The chapters summarize years of extensive research in each area, andprovideinsightinthenewthemesofnaturalproductresearch.Manyofthe contributors illustrate their laboratory experiences. It's obvious that readers will gain exciting and essential information fromthe volume. In the ?rst chapter, Kayser et al. describe the chemistry, biosynthesis and biological activities of artemisinin, one of the most promising antimalarial molecules, and its related natural peroxides. They present new strategies of producing artemisinin that utilize fascinating technologies. as Additionally, thepharmacokineticpro?leandthedevelopmentofnewdrugdeliverysystems onPlasmodium infected erythrocytesare presented . Khan describes some aspects of sugar-derived heterocycles and their p- cursors which are utilized as the inhibitors against glycogen phosphorylases (GPs), and are responsible for the release of mono-glucose from poly-glucose (glycogen) in the second chapter. The inhibitors of GP could help to stop or slowdownglycogenolysisaswellasglucoseproduction.Ultimately, thewhole processwill result in the recovery fromdiabetes of NIDDMpatients. In his contribution, Verma studies quantitative structure-activityrelati- ship (QSAR) and proposes several interesting QSAR models of heterocyclic molecules having cytotoxicactivities against different cancer cell lines, which couldin turn clarify the chemical-biologicalinteractions ofsuch compounds. Alamgir et al., in the fourth chapter, review the recent progress of the synthesis, reactivity and biological activities of benzimidazoles. Additionally, they describe several new techniques and procedures for the synthesis of the same scaffold. Inthe?fthchapter,KhanreviewstheessentialroleoftheenzymeTyrosinase in human melanin production, covering various related clinical problems.

GTPases in Biology II
The GTPase switch appears to be almost as old as life itself, and nature has adapted it to a variety of purposes. This two-volume work surveys the major classes of GTPases, including their role in ensuring accuracy during protein translation, a new look at the trimeric G-protein cycle, the molecular function of ARF in vesicle coating, the emerging role of the dynamin family in vesicle transfer, GTPases which activate GTPases during nascent protein translocation, and the many roles of ras-related proteins in growth, cytoskeletal polymerization, and vesicle transfer. 80 chapters contain much previously unpublished data and, at the rate the extended family of GTPases is growing, it is unlikely that it will again sit for a group portrait such as this. Thus, this could well become the standard reference work.

Endocytosis in Plants
Endocytosis is a fundamental cellular process by means of which cells internalize extracellular and plasma membrane cargos for recycling or degradation. It is important for the establishment and maintenance of cell polarity, subcellular signaling and uptake of nutrients into specialized cells, but also for plant cell interactions with pathogenic and symbiotic microbes. Endocytosis starts by vesicle formation at the plasma membrane and progresses through early and late endosomal compartments. In these endosomes cargo is sorted and it is either recycled back to the plasma membrane, or degraded in the lytic vacuole. This book presents an overview of our current knowledge of endocytosis in plants with a main focus on the key molecules undergoing and regulating endocytosis. It also provides up to date methodological approaches as well as principles of protein, structural lipid, sugar and microbe internalization in plant cells. The individual chapters describe clathrin-mediated and fluid-phase endocytosis, as well as flotillin-mediated endocytosis and internalization of microbes. The book was written for a broad spectrum of readers including students, teachers and researchers.

Noise-Induced Transitions: Theory and Applications in Physics, Chemistry, and Biology
The study of phase transitions is among the most fascinating fields in physics. Originally limited to transition phenomena in equilibrium systems, this field has outgrown its classical confines during the last two decades. The behavior of far from equilibrium systems has received more and more attention and has been an extremely active and productive subject of research for physicists, chemists and biologists. Their studies have brought about a more unified vision of the laws which govern self-organization processes of physico-chemical and biological sys tems. A major achievement has been the extension of the notion of phase transi tion to instabilities which occur only in open nonlinear systems. The notion of phase transition has been proven fruitful in apphcation to nonequilibrium ins- bihties known for about eight decades, like certain hydrodynamic instabilities, as well as in the case of the more recently discovered instabilities in quantum optical systems such as the laser, in chemical systems such as the Belousov-Zhabotinskii reaction and in biological systems. Even outside the realm of natural sciences, this notion is now used in economics and sociology. In this monograph we show that the notion of phase transition can be extend ed even further. It apphes also to a new class of transition phenomena which occur only in nonequilibrium systems subjected to a randomly fluctuating en vironment.
