The cyclic purine nucleotides 3',5'-cAMP and 3',5'-cGMP are well-established second messengers. cGMP has recently been covered in a volume of the Handbook of Experimental Pharmacology (volume 191). In addition to 3',5'-cAMP and 3',5'-cGMP, so-called non-canonical cyclic nucleotides exist. These comprise the cyclic pyrimidine nucleotides 3',5'-cCMP and 3',5'-cUMP, the purine nucleotide 3',5'-cIMP, the 2',3'-nucleoside monophosphates and cyclic dinucleotides. In this volume of the Handbook of Pharmacology, word-leading experts in the field summarize our current knowledge on these non-canonical cyclic nucleotides, discuss open questions, future research directions and the pharmacotherapeutic implications. Special emphasis will be given to the emerging roles of 3',5'-cCMP and 3',5'-cUMP as second messengers with regard to generators, effectors, biological functions, inactivation and bacterial toxins. The role of 3',5'-cIMP as potential second messenger will also be critically discussed. Furthermore, we will consider transport of cyclic nucleotides and their potential role as first messengers. The role of the cyclic dinucleotide cGAMP in the immune system will covered, too. It discusses our current understanding of the molecular mechanisms of DOHaD, including gene body epigenetics and non-coding RNA, and comprehensively examines diseases such type 2 diabetes, a well known as standard DOHaD-associated disease, as well as non-alcoholic fatty liver disease, hypertension and neurodevelopmental disorders. It argues that most adult diseases start at a very early stage, such as in the fetal and neonatal periods, and that earlier prevention and intervention would result in better outcomes for adult diseases such as type 2 diabetes and cardiac disorders, which are increasing in both developed and developing countries. This volume reviews recent advances concerning the glucose-sensing receptor in pancreatic beta-cells. The history of research into pancreatic beta-cells is long and complex; accordingly, the first chapters present the history of this field and explain the hypothesis of insulin secretion mechanisms: glucoreceptor theory . Early Nutrition and Lifestyle Factors: Effects on First Trimester Placenta
This book highlights the impact of nutrients on early placentation processes and their relevance for fetal growth and pregnancy outcome. The role of maternal nutrition on fetal growth and development has been evidenced in many epidemiological studies that included infamous Dutch famine, Helsinki Birth cohort and others. Fetal programming hypothesis states that the nutritional and other environmental conditions under which an individual develops from pre-conception to birth has a major impact on the future health of the newborn child. The developmental environment of the fetus is primarily dependent on two major factors that are maternal nutritional state (excess/low/imbalance) and placental function. Placentation is characterized by the extensive remodeling of the maternal uterine vasculature producing low-resistance blood vessels that facilitate the exchange of nutrients and wastes between the mother and the fetus. Cellular and molecular mechanisms involved in human placental blood vessel formation, which are now well established, are discussed.
